Human ADME-Tox
The Plexpress Human ADME-tox library provides a simple and flexible way to measure gene expression levels of the CYP (Cytochrome P450) -encoding genes and other relevant tox genes associated with drug metabolism from a single sample. In comparison to RT-qPCR, TRAC (TRanscriptional Analysis with the aid of affinity Capture) will allow quicker, more reliable and cost-efficient measurement of mRNA in hepatocytes or HepaRG cells.
Technical note: Human cytochrome P450 expression screening in primary hepatocytes
The Plexpress Human CYP panel offers the followings advantages over alternative methods:
- Direct measurement from crude cell lysates cultured in 96-well plates
- No RNA extraction or cDNA conversion required
- Degradation of RNA avoided, high reproducibility with CVs below 10%
- Multiple housekeeping genes available for use in the same assay (ACTB, SDHA, B2M, GAPDH, UBC, HPRT, RPL13A)
- The flexibility of TRAC enables easy customization of the panel and the addition of other genes of interest
